1st fret tuning and intonation? Check your guitar to see if your 1st fret fretted notes are in tune. Most are out, if you fret a low E string on 1st fret in standard tuning it should register F. Most go towards F#.

Change strings regularly to play and stay in tune. Stretch them out while connected to tuner and stretch until they don't change or go flat. Find your gauge of string and stick with it each time you change & buy. Wipe your strings off after playing. Have a pro set-up your guitar to your liking. Take care of your instrument so it will last longer.
Keep your acoustic guitar in a good place, not in direct sunlight by the window. guitars /fingerboards and necks dry out due to our naturally harsh dry conditions. Change strings regularly and moisten finger board as needed. Get a humidifier for your guitar room if possible.
Clean your bass strings after playing and they will last a lot longer. Check your intonation for the current tuning you are using mostly. Standard or half-step down. Check your intonation on your first fret to make sure your nut is cut correctly and in tune.
Start with everything at halfway up (12 o'clock) except master volume. Turn off all built-in effects off at this time. Depending on clean or dirty sound add gain/drive into sound. Dial your master up until it's too loud, then back off a bit that everyone can live with. Then slowly introduce any effects you are going to use modestly.
FX should always be thought of as an addition to your existing sound or tone. You never want the effects to take over your dry signal. So always run your effects low at first and then ease them into your sound. Effects loops are always best for reverbs/delays on guitars. Make sure you have correct power adaptor and it's not humming or buzzing because of incorrect voltage/current.
Check all your gear early the day before or the of your gig, make sure you have spare strings, cables and anything that is crucial. Best to pack the night before leaving. Have a spare or back-up plan for amps/modelers/crucial pedals if possible. Small amp heads are cheap and a great save. Check your cable, pedalboard, guitars/basses and amps out like you're playing the show or rehearsal.
